- Ran gels of yesterday's digest
Not exactly band patterns we were hoping for. Still looks like uncut DNA. However some do have bands.
Troubleshooting: NEB website says no cutting may be because of too high a ratio of DNA solution to others (Miniprepped DNA has salt, which interferes with restriction enzymes. Miniprep concentrations were around 50ng/ul)
- Did digests again on select pENTRs
- 40ul total volume instead of 20ul, same amount DNA (500ng)
- Ran gel
