Remember:
If you want sequencing to be picked up that day, your samples MUST be in the dropoff box on the first floor of 68 by 3PM. The box is located near the DHL and FedEx mailboxes near the main stairs.
Samples must be prepared in 8 strip PCR tubes. If you’re using less than 8, cut off the empty tubes for future use.
Preparing your samples:
In the sample tube, mix 10 ul of your template (at specified concentration - see below for plasmids vs. pcr products) and 5ul of your primer.

Genewiz has a list of universal primers that your vector may have and they will add these for you at no added cost. If you choose to use these, only add 10ul of your template.
Before submitting, you must place your sequencing order online. The online ordering system will create tube labels using your initials and the sample number. Use these labels on your tubes.
Print a copy of the order receipt and send it along with your samples in an sealed and labeled envelope.
Additional Information from Genewiz Regarding Turnaround
** Highlights are that you should receive results the next day and you can call their Cambridge lab if you don't. Also, premixed samples are cheaper so if you can do that, it's helpful. **
In Boston, we do not charge extra for same day sequencing. You don't need to mark them samples as same day to get the same day service. Apparently, Genewiz took that option off the form as it applied to all samples in Boston anyway.
When you put your samples in the drop box by 3PM you will get your results by 1PM the next day for the same price. ($6 per reaction for premixed samples submitted in .2ml tubes or $4 per reaction for 48 or more premixed samples submitted in 96-well plates.) If you miss your courier pickup we accept samples at the door until 6:30pm. If you need your samples before 1pm for a special project - then you can call the lab and ask them to put your samples on one of the first runs of the day. For Predefined - which is when you adjust the DNA and your primers and have us mix them together or we are storing your primer for you we charge an additional $1 per reaction. If you send in bacterial colonies or other custom samples (you have us adjust your DNA and primers) then we charge and additional $4 per reaction for less than 48 samples or an additional $2 per reaction for 48 or more reactions per order. In other words, for bacterial colony sequencing where we sequence directly off the colonies - we charge $10 per reaction for less than 48 colonies per order or $6 per reaction for 48 or more colonies per order. We consider this a great value as you don't have to do a mini prep! We use a rolling circle amplification process so it requires an additional 12 hours for sequencing but it is often it as you can be doing other things then mini preps! You don't get DNA back but you do get a very clean sequence result. It will be slightly shorter 600 to 700bp vs 850bp from a plasmid - but again it could be more valuable to you.
